"The Lizard" is not a villain. He isn't even the product of callous hubris. He's a mishap. If he is the "villain" of the next Spider-Man movie, he will mar the movie in the same way that Mr. Freeze ruined "Batman and Robin". I am similarly unenthusiastic about the presence of Carnage. Electro would be better suited to be the primary adversary.

Here´s my speculation and I think it will be like this, we all know of the bad response Spider-Man 3 got, and now there´s been talk of a reboot for the Spider-Man franchise after Spider-Man 4. That means that you actually have to most likely go all in for Spider-Man 4. One of the main subjects in the Spider-Man movies have been the villains. And what better way to do that than having Lizard as a villain who practically has been prepared the latest Spider-Man movies. Bruce Campbell will also probably play the role as a villain, for starters he has been in some of the Spider-Man movies. Yeah he will probably be in Spider-Man 4 as a villain. Go all in!
